The parents of Sirianni are Fran and Amy. He is of Italian descent. Nick graduated in 1999 from Southwestern Central High School in West Ellicott, New York, where his father served as the head coach.

At Division III Mount Union in Alliance, Ohio, Sirianni was a wide receiver who helped the Mountaineers win 2000, 2001, and 2002 national titles. Sirianni started for three years even though a calf injury and compartment syndrome almost put an end to his playing career as a sophomore.
He finished his senior year with 998 yards and 13 touchdowns, and also a degree in education. He participated in one season of the American Indoor Football League competition for Canton Legends.
Under him, the 2022 Eagles finished the regular season with a 14-3 record, winning the NFC East division and a bye in the first round of the playoffs. The Eagles set a franchise record for victories and points scored in a season and were the first team since the 1989 Minnesota Vikings to record at least 70 sacks.
They also lead the league in the fewest passing yards allowed. After defeating the New York Giants 38-7 in the divisional round and the San Francisco 49ers 31-7 in the NFC Championship, Sirianni guided the Eagles to their fourth Super Bowl in team history.
