Walker, who was born in Thomaston, Georgia, on December 18, 2000, attended Upson-Lee High School. He was chosen for the All-American Bowl in 2019. He chose to play college football at his native Georgia University in spite of getting offers from other universities.

Walker shed 15 pounds and enjoyed a breakthrough season with the Bulldogs as an edge rusher who could still play inside after two seasons as a backup. He recorded 6 sacks, 33.5 tackles for a loss, and 33.5 total tackles. He played in nine games as a reserve the following season after being chosen to the Freshman All-SEC Team in his first season in Athens with 15 tackles, 3.5 for loss, and 2.5 sacks in 12 games (13 tackles, two for loss with one sack). For the 2021 national champions, he made all 15 starts, notching 37 tackles, 7.5 sacks with six starts, and two pass breakups.

Walker was a member of the squad that won the 2022 College Football Playoff National Championship and made his defensive tackle debut as a junior. After the season, he declared for the NFL Draft in 2022.

Still, not many NFL Draft mocks included Walker’s ascension to a potential first-round selection. After all, Georgia’s defense included All-American players in defensive tackle Jordan Davis, linebacker Nakobe Dean, and safety Lewis Cine at every level.

The Jacksonville Jaguars made Walker their first-round pick in the 2022 NFL Draft. On May 12, 2022, he agreed to a four-year rookie deal worth $37.4 million that was completely guaranteed. Walker was the first athlete taken No. 1 since Matthew Stafford in 2009 and the fifth player from Georgia to do so.
